1-Benzyl-4-methylpiperazine dihydrochloride (1BMP) is a structural analog of imatinib, which is an allosteric inhibitor of the enzyme b-raf. 1BMP binds to the ATP binding site on b-raf and prevents it from activating its target protein, the kinase enzyme that plays a key role in cell division. Imatinib has been shown to be effective in treating chronic myeloid leukemia and other cancers with mutations in the b-raf gene. Sorafenib, another allosteric inhibitor of b-raf, was also found to be effective against chronic myeloid leukemia, as well as pancreatic cancer and hepatocellular carcinoma.
