AS 2444697 is a small molecule that inhibits the inflammatory cytokine TNF-α. It has been shown to inhibit the production of other proinflammatory cytokines, such as IL-1β and IL-6. AS 2444697 also has a protective effect on chronic kidney disease by decreasing necrotic cell death. This drug also inhibits the synthesis of inflammatory mediators such as prostaglandins and leukotrienes by inhibiting cyclooxygenase (COX) and lipoxygenase enzymes. AS 2444697 also inhibits cancer growth by blocking toll-like receptor 4 (TLR4) signaling, which is important in tumorigenesis. It binds to the TLR4 receptor, preventing activation of downstream molecules including NF-κB and MAPKs that are involved in tumorigenesis. AS 2444697 has been shown to have anti-inflammatory effects in an animal model of endometriosis with lower doses than those used in cancer treatment.
