Cidofovir is an antiviral drug that is used in the treatment of cytomegalovirus (CMV) and other viral infections. It inhibits viral DNA synthesis by competing with deoxyguanosine triphosphate as a substrate for viral DNA polymerases, which results in the accumulation of incomplete chains. Cidofovir also has significant cytotoxicity against infected cells. Cidofovir is administered orally as a prodrug and is converted to its active form in the liver. The prodrug is then phosphorylated to cidofovir diphosphate and triphosphate, which are incorporated into the growing DNA chain and inhibit further progression of the chain by competitively inhibiting the activity of DNA polymerase.
