1,4,7,10-Tetraazacyclododecane-1,7-diacetic acid (DOTA) is a chelating agent that forms stable complexes with a range of metal ions. It is used as a radiotracer for positron emission tomography (PET) and single photon emission computed tomography (SPECT). DOTA has been shown to accumulate in cancer tissues and organs due to its high molecular weight. DOTA has also been used as an anti-inflammatory compound and it has been found to inhibit the release of pro-inflammatory cytokines. Radiolabelled DOTA is commonly used in nuclear medicine diagnostic imaging procedures because it has high uptake in many cancers, including prostate cancer. The reaction mechanism of DOTA involves the formation of an acid from carboxylates. DOTA is metabolized by mammalian cells and can be excreted through urine or bile.
