Veliparib is a pan-inhibitor of polymerase chain reaction (PCR) that inhibits the enzyme DNA polymerase. It has been shown to induce neuronal death and inhibit cell growth in vitro, as well as to reduce tumor size in vivo. Veliparib binds to the active site of DNA polymerase, thereby inhibiting its activity. The binding mechanism is competitive with respect to ATP and is reversible. Veliparib has been shown to be an effective inhibitor of Jak2 in v617F cells, which are found in about 15% of all solid tumours. Veliparib also inhibits the expression of proteins involved in cellular proliferation, such as cyclin D1 and c-Myc, which are associated with increased cancer risk.
