4-Nitrophenyl-β-D-lactopyranoside is the glycosidic form of the natural pentose sugar, pentosan. It has been shown to inhibit bladder cancer cells. 4NP binds to a specific ligand on the surface of bladder cancer cells, which is not present on other cell types, and blocks the activation of an enzyme that synthesizes a sugar called pentosan. This leads to decreased production of pentosan, which is important for tumor growth and survival. The inhibition constant (Ki) for 4-nitrophenyl-β-D-lactopyranoside was determined using a synthetic substrate with known Ki values. A titration assay was used to determine the Ki value for 4NP in human plasma by measuring changes in pH as an indicator of glycosidase activity. The Ki value for 4NP was found to be 0.06 μM.
