Ethyl (4-chlorophenyl)(hydroxy)acetate is a chiral, enantioselective, catalytic, and efficient synthetic reagent for the arylation of ketones. It has been used as an intermediate in the synthesis of various pharmaceuticals. This compound is prepared by reacting ethyl 4-chloroacetate with hydroxylamine and glyoxal in the presence of an acidic catalyst. A mutant of this compound has been developed that contains phosphite groups instead of chlorides to avoid potential side reactions with metal catalysts.
