Toluene-4-sulfonic acid 2-cyclopropyl-ethyl ester (Xen445) is a novel, orally bioavailable and potent inhibitor of lipase. It has been shown to inhibit the activity of both intestinal and hepatic lipases in vivo with no appreciable effect on other proteases. Xen445 is a member of the class of compounds that have been shown to inhibit atherosclerosis by decreasing cholesterol levels and lowering lipid concentrations in atherosclerotic plaques. In mice, Xen445 treatment significantly lowered plasma levels of total cholesterol, low density lipoprotein cholesterol, apolipoprotein B, triglycerides and phospholipids. This agent also has cardiovascular effects in mice through inhibition of nitric oxide production by endothelial cells.
