Methyl 4-chloronicotinate is a potent antifungal agent that is used for the treatment of fungal infections. It has been shown to be effective against many types of fungi, including Candida species, Cryptococcus neoformans, Aspergillus fumigatus, and dermatophytes. Methyl 4-chloronicotinate also exhibits potent antifungal activity against Aspergillus nidulans and Coccidioides immitis. The compound was discovered in 1978 from the fungus Micafungin. It has a linear structure with two terphenyl chains. Methyl 4-chloronicotinate binds to ergosterol in the fungal cell membrane and inhibits protein synthesis by inhibiting echinocandin's ability to bind to the beta-subunit of RNA polymerase. This leads to decreased production of proteins vital for cell division and ultimately cell death.
