5'-Hydrazino-5'-deoxyguanosine is a novel diphosphate nucleoside analog that has been shown to be an efficient activator of the anti-cancer enzyme ribonucleotide reductase. It is synthesized by reacting 5'-hydrazinouridine with hydroxylamine and then coupling with 5'-deoxyguanosine. The resulting product has been shown to inhibit DNA synthesis in vitro and in vivo, as well as to produce cytotoxic effects in human cancer cells. This compound also inhibits bacterial growth by binding to DNA-dependent RNA polymerase, thereby preventing transcription and replication. The high frequency of human activity has been shown using a patch-clamp technique on human erythrocytes.
