Thiophene-2-carboxamide is a nicotinic acetylcholine receptor (nAChR) antagonist that is selective for the α7 nAChR subtype. It inhibits the rate of neurotransmitter release at synapses by binding to and blocking the nAChRs. Thiophene-2-carboxamide has been shown to inhibit the activation of toll-like receptors, which are inflammation markers on cells, in human macrophages. This drug also has serotonergic activity and can act on serotonin receptors, potentially leading to adverse effects such as headache, nausea, vomiting, or diarrhea.
