Grb2 is a member of the Sos family of guanine nucleotide exchange factors. It activates Ras by exchanging bound GDP for GTP. Grb2 also binds to SHP-1 and SHP-2, which are phosphatases that dephosphorylate phosphotyrosine residues on the surface of activated receptors. The GST tag may be used as an affinity tag to purify recombinant Grb2 in E. coli. The recombinant protein has been shown to inhibit protein interactions with the Nck and Shc proteins, which are activators of Ras. Recombinant Grb2 has also been shown to activate Ras independently of its binding to other proteins, such as Shc or Sos, and can bind to a variety of ligands including receptor tyrosine kinases, ion channels, and antibodies.
