5-Bromo-4-oxo-pentanoic acid is an irreversible enzyme inhibitor that targets the lysosomal aspartyl proteases, such as cathepsin B. It is synthesized from canagliflozin and a sulfoxide reagent by an acylation reaction with a tetrapyrrolic compound. 5-Bromo-4-oxo-pentanoic acid inhibits the lysosomal enzyme cathepsin B, which is involved in the degradation of proteins in lysosomes. This drug binds to the active site of cathepsin B and prevents it from hydrolyzing its substrate, thereby blocking protein degradation. 5-Bromo-4-oxo-pentanoic acid also has been shown to inhibit other enzymes, such as lipoprotein lipase, sphingomyelinase, and phospholipase A2.
