4-Amino-2',4'-dichlorodiphenyl ether (4AD) is a diphenyl ether that has been shown to be an activator of the Ah receptor and thus may have potential as a therapeutic agent for the treatment of lung cancer. The mutagenicity of 4AD in a variety of tissues, including human embryonic lung cells, was confirmed in the Ames test. 4AD has also been shown to inhibit the growth of bacteria in emulsions. In long-term studies, 4AD induced teratogenic effects in rats. Pregnant female rats were given doses of 20 and 40 mg/kg/day during gestation days 6 through 15. A statistically significant increase in fetal malformations was observed at both doses. The type and severity of malformations did not appear to be dose related. The incidence and severity were similar to those seen with other known teratogens such as 3,3',4,4'-tetrachlor
