3-Azabicyclo[3.2.1]octan-8-one is a repressor of transcription that belongs to the family of mycobacterial transcriptional strategies. This compound is a prodrug that can be metabolized by cytochrome P450 enzymes in the liver to ethionamide, its active form. 3-Azabicyclo[3.2.1]octan-8-one has been shown to have an additive effect with ethionamide for boosting the activity of tuberculosis drugs against Mycobacterium tuberculosis and Mycobacterium avium complex infections in mice, although it does not show any activity on its own against these strains. The crystal structure of this compound bound to DNA shows that it interacts with the major groove, which may explain its ability to inhibit DNA synthesis and transcription.
