Formin is a protein that is involved in the cytoskeletal regulation of cell shape and motility. The formin FH2 domain inhibitor (SMIFH2) is a potent, selective, and reversible inhibitor of formin-mediated actin polymerization. SMIFH2 binds to the formin FH2 domain and prevents it from binding to other proteins such as myosin IIA. This inhibits the assembly of actin filaments, leading to the inhibition of cellular processes such as leukocyte recruitment, nucleation, and cytochalasin B-induced neutrophil chemotaxis. SMIFH2 has been shown to inhibit neutrophil migration in vitro by preventing the formation of an actin network around the neutrophils.
SMIFH2 also blocks tnf-α induced leukocyte recruitment by inhibiting cytoskeletal reorganization and subsequent cell migration.
