Piperidine-1-carboximidamide hydrochloride (PCI) is an antimalarial agent that belongs to the group of pyrimidines. It inhibits the growth of plasmodium by inhibiting the production of pyrimethamine in the parasites erythrocytes. PCI has been shown to be active against both chloroquine-sensitive and resistant strains of plasmodium falciparum, with a median inhibitory concentration IC50 of 0.8 μM. This drug also has a high level of activity against other species such as Plasmodium vivax and Plasmodium ovale. PCI may act by blocking the conversion of dihydrofolate to tetrahydrofolate, which is required for DNA synthesis and protein synthesis in the parasite.
