5-Chloro-2-nitrobenzoyl chloride (5CNCl) is an antihistaminic agent that has been shown to have analgesic, anticholinergic, and psychoneurotic effects in animal studies. 5CNCl suppresses the synthesis of histamine by inhibiting the enzyme histidine decarboxylase. 5CNCl also inhibits cholinesterase activity, which leads to increased acetylcholine levels in the synapse. In addition, it inhibits the cycloadditions of prostaglandin and thromboxane A2. The synthetic methods for 5CNCl involve reacting 3-chloroaniline with nitroethane and sodium nitrite in a mixture of ethanol and water to produce 5-chloro-2-nitrobenzoic acid. This acid is then reacted with phosphorus pentachloride to form 5-chloro-2-nitrobenzoyl chloride.
