WIN 54461 is a potent and selective agonist of the alpha2-adrenergic receptor. WIN 54461 binds to the alpha2-receptor with high affinity and selectivity, and activates the receptor in a dose dependent manner. WIN 54461 has been shown to inhibit the activity of ion channels such as those for Na+, K+, Ca++, and Cs+ in a variety of tissues including rat brain synaptosomes, guinea pig ileum and rabbit muscle. The effects on ion channels are similar to those seen with clonidine. In addition, WIN 54461 has been shown to antagonize the effects of anesthetic agents such as pentobarbital on neuronal excitability in rat brain slices. WIN 54461 also possesses significant binding affinity for other G protein coupled receptors including dopamine D1, D2, and D3 receptors.
