Product Information
Nornalmefene is a synthetic opioid receptor antagonist that blocks the binding of opioid drugs to the mu and kappa receptors. It is used in the treatment of alcohol dependence. The pharmacokinetic properties of nalmefene are similar to other oral medications, with a half-life of approximately 10 hours. Nornalmefene has been shown to be safe and effective in humans when taken orally and has potential use as a chronic oral therapy for alcoholism. Nalmefene binds to the mu and kappa opioid receptors, which are involved in pain signaling pathways, modulating the effects of opioids such as morphine. Nalmefene also decreases craving for alcohol by inhibiting dopamine release in the nucleus accumbens, an area of the brain involved in addiction.
Nalmefene undergoes metabolism in vivo by CYP3A4 and CYP2D6 enzymes into its active form naloxone, which is responsible for its antagonistic effects on opioid receptors. Active metabolites are
