GBR 12783 is a dopamine uptake inhibitor that is being developed for the treatment of autoimmune diseases and for the prevention of osteoporosis. GBR 12783 has been shown to inhibit dopamine uptake in rat brain synaptosomes and in cultured human cells. This drug has also been found to inhibit the synthesis of glutathione ethyl ester, which could be a potential mechanism for its biological properties. GBR 12783 binds to neurons, causing them to die by irreversible inhibition of gamma-aminobutyric acid (GABA) receptors. It also blocks the binding of GABA to its receptor, causing neuronal death. GBR 12783 has not yet been tested in humans and is not FDA approved.
