2-Hydroxy-5-(methylthio)benzaldehyde (HMB) is a ligand that binds reversibly to a copper ion in the active site of many enzymes. It has been shown to react as a bidentate ligand, forming a tripodal complex with two axial and two equatorial coordination sites. HMB also reacts with perchlorates, forming an ionic complex that is soluble in organic solvents such as dichloromethane or chloroform. The protonation of HMB at the phenolic hydroxyl group leads to its formation of an ionic complex with the axial Cu(I) ion. This reaction is favored by low pH, which causes deprotonation of the hydroxyl group. The reaction can be observed by electrochemical studies.
