2-Chloro-4-cyanobenzoic acid is an electron receptor antagonist that binds to the androgen receptor. It has been shown to be a potent androgen receptor antagonist in vitro and in vivo, with a synergistic effect when combined with other androgen receptor antagonists. This antagonistic activity may be due to the substitution of adamantyl residues in the ligand binding domain of the androgen receptor. 2-Chloro-4-cyanobenzoic acid has also been shown to have an antiestrogenic effect by blocking estrogen receptors. Molecular modeling studies suggest a number of ways for 2-chloro-4-cyanobenzoic acid to bind to the androgen receptor, including docking on top of helix 12 or 17, or through hydrogen bonding with polar groups such as serine residue 587.
