Donepezil is a cholinergic that binds to the acetylcholinesterase (AChE) enzyme, which is involved in the breakdown of acetylcholine. This drug has been shown to be effective in alleviating symptoms of Alzheimer's disease and other cognitive disorders associated with dementia. Donepezil also slows down the progression of Alzheimer's disease by preventing neuronal death. It is metabolized through a number of metabolic transformations, including hydrolysis by esterases or glucuronidases, oxidation by cytochrome P450 enzymes, reduction by glutathione reductase, or conjugation with glucuronic acid. Donepezil also specifically binds to markers expressed at high levels in Mycobacterium tuberculosis strains (e.g., ESX-1 secretion system protein) and inhibits cell growth in culture.
