Bepristat 1a is a small molecule that binds to the active site of the endoplasmic reticulum (ER) enzyme histidine kinase, thereby inhibiting its activity. Bepristat 1a is activated by radiation and reversibly damages DNA in vitro and in vivo. The drug also inhibits tumor growth in vivo without affecting healthy tissues. The drug has been shown to selectively induce apoptosis in cancer cells by inhibition of ERK1/2 phosphorylation and activation of p38 MAPK.
