3,4-Dihydro-2H-1-benzopyran-6-sulfonamide (DHBS) is an orally active retinoid prodrug that is metabolized in the liver to the active form, all-trans retinoic acid. The pharmacokinetic profile of DHBS and its metabolites has been evaluated in rats. The pharmacokinetic parameters of DHBS and its metabolites were determined after oral administration at doses of 10, 20, and 40 mg/kg. The systemic exposure to DHBS was found to be dose-dependent with a peak concentration of 5.3 μM/L at the highest dose administered. The bioavailability was also found to be dose dependent with a bioavailability of 0% at the lowest dose administered but increasing up to 100% at the highest dose administered. The pharmacokinetic profile of DHBS and its metabolites has been evaluated in humans as well. This prodrug showed low systemic exposure when administered orally due
