Th-302 is a phosphorodiamidate prodrug that is converted to methyl N,N'-bis(2-bromoethyl)phosphorodiamidate (th-302) in the body. Th-302 inhibits the growth of solid tumours by interfering with DNA replication and protein synthesis. It has been shown to be effective against some strains of cancer cells that are resistant to radiation therapy. Th-302 has also been shown to inhibit locomotor activity in mice, which may be due to its ability to produce hypoxia and induce hypoxic conditions in the tumor environment. The drug has no effect on healthy tissue because it does not cross the blood-brain barrier. Th-302 also has a hydrophobic effect, which may cause it to accumulate near the outer membrane of cells and prevent oxygen from entering, leading to cell death.
