4-Amino-2-(trifluoromethyl)pyridine-3-carboxylic acid (4ATPC) is a synthetic compound that has been synthesized by lithiation, iodination, and synlett reactions. 4ATPC has been shown to be an effective quencher of carbonyl compounds. It can be used in the synthesis of amines and carboxylic acids. The deprotection of the tertiary butyl group is achieved with sodium hydroxide in methanol or with hydrogen gas in tetrahydrofuran at 0°C. The carboxylation reaction is performed using sodium carbonate and ammonium chloride in methanol or using potassium carbonate and ammonium chloride in ethanol. Lithiated 4ATPC can be prepared by reacting it with lithium metal in ether at -78°C followed by amination with ammonia.
