Benzyl 2-hydroxy-2-methylpropionate is an enzyme inhibitor that inhibits HIV by preventing protease from cleaving the viral polyprotein. It is synthesized in a two-step process, which involves cyclopropanation of benzyl alcohol with ethyl bromoacetate followed by hydrolysis to form the ester. The compound has been shown to inhibit HIV-1 and HIV-2 strains, as well as tipranavir. It is structurally analogous to other protease inhibitors such as cinnamyl and tetronic acids. Benzyl 2-hydroxy-2-methylpropionate binds at the active site of the enzyme and blocks access for substrate, thereby inhibiting cleavage of viral proteins into functional domains. This inhibition leads to decreased synthesis of viral proteins, which can lead to death of cells infected with HIV.
