4-Chloroquinoline-3-carbonitrile is an intermediate in the synthesis of the tyrosine kinase inhibitor pelitinib and the process for its synthesis has been described. The synthetic process starts from acetonitrile, which reacts with nitrous acid to produce 4-chloroquinoline-3-carbonitrile. The next stage involves the reaction of 4-chloroquinoline-3-carbonitrile with ethyl acetate in the presence of a catalyst to produce 2-(4'-chlorophenyl)ethyl acetate. This compound can then be reacted with phenylethylamine to form (2E)-N-[2-(4'-chlorophenyl)ethyl]-N-(2,6-dimethoxyphenyl)acrylamide, which is a key intermediate in the synthesis of pelitinib.
