(2Z)-2-Cyano-N-(2,5-dibromophenyl)-3-hydroxy-2-butenamide is a potent inhibitor of Toll-like receptor (TLR) signaling that can be used to treat bowel disease. It has been shown to inhibit TLR signaling pathways in human monocytes and myeloma cell lines. This drug also inhibits the production of proinflammatory cytokines by inhibiting the nuclear factor of activated T cells (NFAT). It is believed that this drug binds to the DNA binding site on NFAT, preventing its transcriptional activity. The potency of this compound is attributed to its hydrogen bonding interactions with the bases in DNA and its ability to disrupt mitochondrial membrane potential.
