Methotrexate is a drug that is used to treat autoimmune diseases, such as rheumatoid arthritis and inflammatory bowel disease. It is also used to treat cancer, including urothelial carcinoma, and some forms of leukemia. Methotrexate inhibits the production of immune cells by binding to folic acid receptors in lymphocytes. Methotrexate-related compound E (MTCE) is a methotrexate analog with immunosuppressive activity. MTCE binds to the folate receptor in a similar manner to methotrexate, but does not inhibit dihydrofolate reductase like methotrexate does. This means that MTCE has lower toxicity than methotrexate because it does not interfere with DNA synthesis or repair as much as methotrexate does.
