4-(Trifluoromethyl)-2-pyridinecarboxamide is a positive allosteric modulator of the mGlu5 receptor. It has been shown to enhance the activation of synaptic transmission, cognition and symptoms in a mouse model of hypofunction. 4-(Trifluoromethyl)-2-pyridinecarboxamide also has a potent interaction with locomotor activity, which can be measured by spontaneous locomotor activity or by rotarod experiments. This compound is orally active and shows significant effects at doses of 0.1 mg/kg (equivalent to 1 mg/kg in humans).
