(+)-Amlodipine

Description:
(+)-Amlodipine is a dihydropyridine calcium channel blocker primarily used in the treatment of hypertension and angina. It is characterized by its ability to selectively inhibit calcium ions from entering vascular smooth muscle and cardiac muscle, leading to vasodilation and reduced blood pressure. The compound is a chiral molecule, with the (+) designation indicating its specific optical activity. Its molecular formula typically consists of carbon, hydrogen, nitrogen, and oxygen atoms, contributing to its pharmacological properties. (+)-Amlodipine is known for its long half-life, allowing for once-daily dosing, which enhances patient compliance. It is generally well-tolerated, with common side effects including peripheral edema, headache, and fatigue. The substance is often formulated as a salt, such as amlodipine besylate, to improve its solubility and bioavailability. As a member of the calcium channel blocker class, it plays a crucial role in cardiovascular therapy, helping to manage conditions related to high blood pressure and chest pain.

  • (R)-Amlodipine

    CAS:
(R)-Amlodipine is a calcium channel blocker that binds to the L-type calcium channels and inhibits the influx of cytosolic calcium, thereby reducing the excitation of ventricular myocytes and decreasing blood pressure. It is also used for the treatment of endometriosis. The (S)-enantiomer has no pharmacological activity and is not used clinically. The mechanism of action for amlodipine involves binding to the cytosolic Ca2+-ATPase pumps, which are responsible for recycling cytosolic Ca2+ back into the intracellular space. As a result, there is an increase in cytosolic Ca2+, which leads to a decrease in excitability of cardiac myocytes, vasoconstriction, and decreased blood pressure. Amlodipine has been shown to be effective against angiotensin II-induced hypertension in rats by blocking efflux pump activity at the blood-brain barrier.
