It is found in high concentrations in the kidney, liver, and pancreas, and is an important cofactor for mitochondrial enzymes that catalyze the oxidation of pyruvate to acetyl CoA. (R)-(+)-a-Lipoic acid has been shown to be effective against cisplatin-induced nephrotoxicity, oxidative injury caused by ischemia–reperfusion injury, diabetic neuropathy and other conditions. The exact mechanism of action of (R)-(+)-a-Lipoic acid is not known but it may be due to its ability to restore mitochondrial membrane potential or inhibit reactive oxygen species production.
