Rifapentine hydrochloride

Description:
Rifapentine hydrochloride is a synthetic antibiotic belonging to the rifamycin class, primarily used in the treatment of tuberculosis, particularly in combination therapy for both drug-sensitive and drug-resistant strains. It exhibits a broad spectrum of antibacterial activity by inhibiting bacterial RNA synthesis through binding to the DNA-dependent RNA polymerase enzyme. This compound is characterized by its relatively long half-life, allowing for less frequent dosing compared to other rifamycins, which enhances patient compliance. Rifapentine hydrochloride is typically administered orally and is known for its good bioavailability. The hydrochloride salt form enhances its solubility and stability. Common side effects may include gastrointestinal disturbances, rash, and potential liver enzyme elevation, necessitating monitoring during treatment. Additionally, it is important to consider drug interactions, as rifapentine can affect the metabolism of various medications due to its influence on cytochrome P450 enzymes. Overall, rifapentine hydrochloride represents a significant advancement in tuberculosis therapy, particularly in the context of directly observed therapy strategies.

    CAS:

Rifapentine is an anti-tuberculosis drug that belongs to the class of rifamycins. It is the most active of the rifamycins for the treatment of tuberculosis. Rifapentine inhibits bacterial growth by binding to DNA-dependent RNA polymerase, thereby preventing transcription and replication. The high frequency of human activity has been shown using a patch-clamp technique on human erythrocytes. This active form is metabolized through a number of metabolic transformations, including hydrolysis by esterases or glucuronidases, oxidation by cytochrome P450 enzymes, reduction by glutathione reductase, or conjugation with glucuronic acid. Rifapentine also specifically binds to markers expressed at high levels in Mycobacterium tuberculosis strains (e.g., ESX-1 secretion system protein) and inhibits cell growth in culture.
