It has been shown to inhibit the endoplasmic reticulum Ca2+-ATPase, which transports calcium ions out of the cell, leading to increased concentrations of calcium ions in the cytosol. 8AADPR also inhibits mitochondrial ATPase, which leads to an accumulation of ADP-ribose within mitochondria. This accumulation leads to the inhibition of protein synthesis within mitochondria and an activation of ryanodine receptors.
