PF 06256142 is a novel, orally active, selective allosteric modulator of the dopamine D2 receptor with functional selectivity. It has been shown to desensitize the dopamine D2 receptor and reduce locomotor activity in animal models. PF 06256142 has also been shown to be effective in clinical studies for the treatment of Parkinson's disease. In vitro assays have demonstrated that PF 06256142 is a potent dopamine D2 receptor allosteric modulator with a binding affinity (Ki) of 0.4 nM and an intrinsic efficacy of about 300%. The compound demonstrates high potency and efficacy in vitro, as well as a favorable pharmacokinetic profile in vivo.
