    <p>The FRET substrate Dabcyl-RGVVNASSRLA-Edans has been synthesized to develop a fluorescence-based assay of human cytomegalovirus proteinase. It is cleaved specifically at the Ala-Ser bond thereby liberating the C-terminal peptide-EDANS fragment from the proximity quenching effect of the DABCYL group. This represents the first fluorescence-based assay of the herpes virus proteases and permits the characterization of potential inhibitors. Cleavage yields the N-terminal fragment DABCYL-RGVVNA.</p>
