    <p>RU.521 is a novel cytosolic inhibitor of autophagy that has been shown to activate the innate immune response and promote chronic pulmonary inflammation in animal models. The molecule binds to toll-like receptor 4 (TLR4) on human macrophages and elicits the release of tumor necrosis factor alpha (TNFα), IL-1β, and IL-6, which are proinflammatory cytokines that promote the activation of dendritic cells. This compound also induces polymerase chain reaction (PCR)-amplified mRNA transcripts encoding TLR4 in primary cells.
