It is a racemic mixture of two enantiomers, (+) and (-) guanfacine. Guanfacine acts on central alpha-2 receptors to reduce sympathetic outflow from the vasomotor center, which leads to reduced peripheral vascular resistance, decreased blood pressure, and increased heart rate. It also has been shown to increase the release of gamma-aminobutyric acid (GABA), a neurotransmitter that inhibits neuronal activity. Guanfacine is metabolized by CYP3A4 and CYP2D6 enzymes with elimination half-life of 10 hours. The drug has minimal drug interactions due to its low protein binding properties.
