It has been shown to inhibit the activity of ryanodine receptors and P-glycoprotein (Pgp) in cells, which are proteins that regulate calcium release from the sarcoplasmic reticulum. Nadolol has also been shown to inhibit the polymerase chain reaction (PCR) and other DNA polymerases, leading to a decrease in the rate of DNA synthesis. Nadolol also decreases blood pressure by inhibiting the effects of catecholamines on myocardial contractility. It binds to α1-acid glycoprotein, thereby reducing its ability to bind with nadolol and increase its plasma concentration. Preparative high performance liquid chromatography (HPLC) can be used for the separation of nadolol from interfering substances.
