Nadolol

Description:
Nadolol is a non-selective beta-adrenergic antagonist, commonly referred to as a beta-blocker, primarily used in the treatment of hypertension and certain types of heart conditions. Its chemical formula is C17H27NO4, and it features a phenolic structure with a secondary amine. Nadolol is characterized by its long half-life, allowing for once-daily dosing, which enhances patient compliance. The substance is soluble in water and exhibits a moderate lipophilicity, which influences its pharmacokinetic properties. Nadolol works by blocking the action of catecholamines on beta-adrenergic receptors, leading to decreased heart rate and myocardial contractility, ultimately reducing blood pressure. It is also utilized in the management of migraine prophylaxis and certain types of anxiety. Common side effects may include fatigue, dizziness, and bradycardia. Due to its pharmacological profile, nadolol is contraindicated in patients with asthma or certain types of heart block. As with all medications, it is essential to use nadolol under medical supervision to monitor for potential interactions and side effects.

    CAS:

Nadolol is a beta-blocker that is used to treat high blood pressure. It has been shown to inhibit the activity of ryanodine receptors and P-glycoprotein (Pgp) in cells, which are proteins that regulate calcium release from the sarcoplasmic reticulum. Nadolol has also been shown to inhibit the polymerase chain reaction (PCR) and other DNA polymerases, leading to a decrease in the rate of DNA synthesis. Nadolol also decreases blood pressure by inhibiting the effects of catecholamines on myocardial contractility. It binds to α1-acid glycoprotein, thereby reducing its ability to bind with nadolol and increase its plasma concentration. Preparative high performance liquid chromatography (HPLC) can be used for the separation of nadolol from interfering substances. Plasma mass spectrometry is often used for confirmation of identity and purity analysis of nadolol preparations.
