CAS:<p>Dantrolene is a drug that has been used for the treatment of muscle spasms, including those caused by tetanus, dystonia, and other neuromuscular diseases. Dantrolene binds to the ryanodine receptor in the sarcoplasmic reticulum of skeletal muscle cells and blocks calcium release from these stores, leading to relaxation of contracted muscles. It also inhibits the uptake of organic anions through P-450 isozymes in the liver and brain. Dantrolene has potent inhibitory activity on p-450 isozymes and gastrocnemius muscle contractility. This drug also has been shown to be effective as a pharmacologic agent for treating renal proximal tubule cells in vitro. Dantrolene can be administered orally or intravenously and its terminal half-life is approximately 2 hours.
