CAS 530-62-1
:1,1′-Carbonyldiimidazole
Description:
1,1′-Carbonyldiimidazole (CDI) is a versatile chemical compound widely used as a coupling agent in organic synthesis, particularly in peptide and ester formation. It features a symmetrical structure with two imidazole rings connected by a carbonyl group, which contributes to its reactivity. CDI is typically a white to off-white solid at room temperature and is soluble in organic solvents such as dichloromethane and dimethylformamide. Its reactivity stems from the carbonyl group, which can activate nucleophiles, making it effective for acylation reactions. CDI is known for its stability under ambient conditions but should be handled with care due to its potential to release imidazole upon hydrolysis, which can lead to unwanted side reactions in sensitive environments. Additionally, it is important to note that CDI can be hazardous, and appropriate safety measures should be taken when handling this compound, including the use of personal protective equipment and working in a well-ventilated area. Overall, CDI is a valuable reagent in synthetic organic chemistry, facilitating the formation of various chemical bonds.

CAS:
CDI is a coupling agent used mainly for amide and ester formation, but also thioesters, ureas and carbamates. CDI is used in a one-pot procedure, where an acyl imidazolide is formed first upon activation of the carboxylic acid with CDI. The amine or alcohol is then added, and it readily reacts with the active intermediate and the product is generated. CDI does not require the presence of base. The addition to the CDI reaction mixture of carbon dioxide or weakly acid compounds such as imidazole hydrochloride, was reported to increase the reaction rates. CDI can be used in large-scale synthesis thanks to its affordable cost, its availability, and the ease of removal of the by-products during the workup, which are carbon dioxide and imidazole. CDI can be employed as a replacement of the poisonous phosgene, as an imidazolium transfer reagent and in the synthesis of lead sequestering agents.
Formula:C7H6N4OColor and Shape:Off-White PowderMolecular weight:162.15 g/mol1,1'-Carbonyldiimidazole (>90%)
