2,4-Dichlorophenylsulfonyl isopropyl (2-((isopropyl)amino)ethyl)amine ((2,4-DAPIS)) is a potent and selective inhibitor of the protein kinase PKCζ. It has been shown to be active in animal models of chronic cough and bronchial asthma. The 2,4-DAPIS binds to the ATP site on PKCζ and inhibits its activity by binding to the catalytic domain of the enzyme. Inhibition of PKCζ leads to decreased angiotensin system activity, which in turn decreases chronic cough and bronchial asthma. The 2,4-DAPIS also blocks potassium channels TRPV4, which may lead to an improvement of disease activity.
