The azido group is sensitive towards piperidine. In Fmoc-SPPS, this derivative is best introduced as N-terminus or modified before elongating the peptide. Fmoc-p-azidophenylalanine may be inserted as a precursor of p-aminophenylalanine. Levinson et al. reduced the azido group on-resin with a large excess of tributylphosphine in DMF.
