Elastase (Pancreatic elastase, elastase-1, pancreatopeptidase E; CAS No [848900-32-3], EC 3.4.21.36) is an enzyme that breaks down elastin, a structural protein that plays a key role in the mechanical properties of connective tissues. It belongs to a serine protease family, as it has a serine in its active site.One unit of elastase will hydrolyze 1.0 μmol of methoxy succinyl-alanine-alanine-proline-valine-p-nitroanilide per minute at pH 7.5 and 37°C. Porcine pancreatic elastase is supplied in lyophilized form. Its activity is ≥1U/mg solid, and specific activity is ≥5U/mg protein. Enzyme is activated by tris and sodium sulfate, optimum pH is 8.5, molecular weight is 25,908Da. Porcine pancreatic elastase's intended uses include digestion of elastin, tissue dissociation, solubilization of membrane proteins and protein structural studies. Store at -20°C on arrival.
