Protoanemonin is a phenolic compound that was isolated from the leaves of the Australian tree Melaleuca alternifolia. It has been shown to have toxicological properties, such as hemolytic activity, and is thought to be responsible for the toxicity of the tea tree oil. Protoanemonin has been shown to inhibit bacterial growth by acting as a competitive inhibitor in fatty acid synthesis, thereby preventing the formation of carbon disulphide. This mechanism may be due to its ability to bind with the enzyme acyl-CoA synthetase and prevent fatty acid chain elongation necessary for bacterial cell wall biosynthesis. Protoanemonin also inhibits other enzymes involved in bacterial growth, such as phospholipase A2, which is responsible for releasing arachidonic acid from cell membranes.
