Triparanol is a drug that inhibits the biosynthesis of cholesterol by blocking the conversion of squalene to lanosterol. Triparanol was originally developed as a contraceptive, but it was abandoned because it caused too many side effects. Triparanol has been shown to disrupt the biosynthesis of cholesterol in animals and humans. It binds to a specific site in the enzyme lanosterol 14α-demethylase and blocks its activity, thereby inhibiting the conversion of lanosterol to desmosterol and 5α-cholestane-3β-ol. In addition, triparanol has been shown to modulate other enzymes involved in cholesterol biosynthesis.
