Hydrocodone is a narcotic analgesic that binds to opioid receptors in the body. It is a semi-synthetic opioid derived from codeine, which is obtained from the opium poppy. Hydrocodone has been shown to have higher affinity for mu-opioid receptors than other opioids such as morphine, and it also has less affinity for kappa- and delta-receptors. Hydrocodone has a concentration-time curve that can be described by three phases: an initial rapid elimination phase, followed by a slower elimination phase with a plateau effect, and finally an elimination phase with decreasing concentrations. Hydrocodone may interact with nonsteroidal anti-inflammatory drugs (NSAIDs) such as aspirin or ibuprofen, since NSAIDs inhibit the enzyme cyclooxygenase 1 (COX1), which is responsible for conversion of arachidonic acid to prostaglandins that produce pain and inflammation.
